You tried turning it on and off again?
(You tried reinstalling windows or running a diagnostic)

Have you tried to restore windows to a earlier restore point. ie: before an update?

Ditch the shampoo, demand the real poo!
Oh dear....
Oh god I'm sorry; this is generally a pain of an issue to fix.
Attempt 1)
When booting up, mash the F8 key to get to a screen that allows you to boot into safe mode. If you are able to boot to this then try a restore to a previous date.
Attempt 2)
Boot to an install disk of the OS that you're running (so Win7 install disk if you are using Win7)
Instead of clicking 'Install Now', click 'Repair your computer', choose your OS, then click 'Command Prompt'.
In command prompt type the following three commands:
bootrec /fixmbr
bootrec /fixboot
bootrec /rebuildbcd
Try booting again.
Attempt 3)
Boot to the command prompt just like the last attempt had you do.
Type the command:
chkdisk C: /r
If none of those work then good luck. We had a client whose computer did this and somehow partitions got moved all over the place and volumes went missing. It was a real mess that we spent like 30 hours trying to fix.

Criminal Lawyer is a redundancy
Happened to me. I got a error for my registry did not have any security files after downloading avg... yup anti virus screwed my system up instead of help.
Lead to me just having to reinstall windows. I did not have a disk at the time so I used a bootable USB and it fixed it just fine fixing the problem. Lost all my files though =/
